چکیده

Of patients with castrate resistant prostate cancer (CRPC), less than 25-33% survive more than five years. Recent studies have implicated estrogen, acting either alone or synergistically with androgens in the development of castrate resistant prostate cancer. Several in vitro and in vivo studies, as well as a limited number of clinical trials, have highlighted the potential of selective estrogen receptor modulators, such as raloxifene (Ral) for the treatment of castrate resistant prostate cancer. However, the poor oral bioavailability and metabolism of selective estrogen receptor modulators limit their efficiency in clinical application. To overcome these limitations, we have used styrene co-maleic acid (SMA) micelle to encapsulate raloxifene. Compared to free drug, SMA-Ral micelles had 132 and 140% higher cytotoxicity against PC3 and DU 145 prostate cell lines, respectively. SMA-Ral effectively inhibits cell cycle progression, increases apoptosis, and alters the integrity of tumor spheroid models. In addition, the micellar system induced changes in expression and localization of estrogen receptors, epidermal growth factor receptor (EGFR), and downstream effectors associated with cell proliferation and survival. Finally, SMA-Ral treatment decreased migration and invasion of castrate resistant prostate cancer cell lines. In conclusion, SMA-Ral micelles can potentially benefit new strategies for clinical management of castrate resistant prostate cancer.

Castrate-tolerant cells: what are the implications for the treatment of localized prostate cancer?

More effective treatment of prostate cancer relies on eliminating cells that survive androgen withdrawal therapy. The discovery that castrate-tolerant tumour cells pre-exist in localized prostate cancer, prior to androgen withdrawal or progression to castrate-resistant disease, supports the notion that neo-adjuvant therapies might be considered in the management of early stage prostate cancer. ...

Guidelines for the management of castrate-resistant prostate cancer.

Advanced prostate cancer has been known under a number of names over the years, including hormone resistant prostate cancer (HRPC) and androgen insensitive prostate cancer (AIPC). Most recently, the terms castrate resistant prostate cancer or castration recurrent prostate cancer (CRPC) were introduced with the realization that intracrine/paracrine androgen production plays a significant in the ...

ETS1 transcriptional activity is increased in advanced prostate cancer and promotes the castrate-resistant phenotype.

Advanced disease accounts for the majority of prostate cancer-related deaths and androgen deprivation therapy (ADT) is the standard of care for these patients. Many patients undergoing ADT become resistant to its effects and progress to castrate-resistant prostate cancer (CRPC). Current therapies for CRPC patients are inadequate, with progression-free survival rates as low as 2 months. The mole...

Castrate-resistant Prostate Cancer— Recent Advances in Therapy and Future Perspectives

For many years, few therapeutic options were available for the treatment of advanced prostate cancer. Recent advances in our understanding of the molecular biology of prostate cancer, particularly in the transition to castrate resistant disease, have led to the development of more potent and selective endocrine therapies. In addition, elucidation of the many factors in the bone microenvironment...

A Systematic Review of Clinical Practice Guidelines for Castration-Resistant Prostate Cancer

Cancer constitutes a huge burden on societies in countries with any level of economic development. Prostate cancer is the first most diagnosed cancer of men in developed countries and the forth one in developing countries in terms of incidence rate. It is also the third incident cancer of men in Iran along with a prevalence of about 10,000 cases. Castration-resistant prostate cancer (CRPC) is a...
